At VHLGenetics we employ various techniques, including but not limited to the following.
DNA extraction
For all genotyping projects, VHLGenetics provides comprehensive DNA extraction services. This service is available both as part of a genotyping project package and as a standalone offering. In the standalone service, clients can submit raw materials such as seeds, leaves or hair, and in return, they receive 96 well plates containing the extracted DNA.
VHLGenetics offers two distinct types of DNA extraction services. The first is tailored for low-end genotyping techniques such as KASP (SNP), fragment analysis (STR, SSR, VNTR), and TaqMan (qPCR, SNP). The second, more advanced protocol, is optimized for sophisticated applications like genotyping-by-sequencing or BeadChips analysis. Additionally, if needed, RNAse treatment and normalization procedures can be applied to the extracted DNA to ensure its quality and integrity.
KASP
The KASP™ (Kompetitive Allele Specific PCR) genotyping assay utilizes a homogeneous fluorescence (FRET) technique for precise bi-allelic discrimination of known SNPs and InDels. Unlike other PCR-based assays, KASP doesn’t need target-specific primer/probe labeling, offering great flexibility in assay design and higher success rates, even for large/mega InDels.
Central to KASP is its use of a universal FRET cassette reporter system, eliminating the need for dual-labeled probes. Each allele-specific forward primer has a tail sequence corresponding to one of two FRET cassettes: FAM dye or HEX dye. This innovation allows KASP to operate in miniaturized reaction volumes (10 µL in 96-well PCR plates, 5 µL in 384-well PCR plates, and 1 µL in 1536-well PCR plates) while maintaining individual PCR reactions.
KASP displays remarkable versatility, capable of accommodating anywhere from 1 to 100 markers.
Genotyping arrays / Genome scan
VHLGenetics offers a comprehensive selection of BeadChips, including both custom arrays and standard Illumina arrays. These arrays feature SNP markers ideal for a variety of genetic applications in both animals and plants.
Our arrays can be employed on a project-by-project basis, providing valuable insights into the genomic background of cattle breeds and populations.
